Characterization of Critical Parameters in Bioprocess Development - Development of a Shaken Scale-Down Model

Characterization of Critical Parameters in Bioprocess Development - Development of a Shaken ...

A shaken scale-down model at approx. 0.1 L scale for a previously established stirred-tank model at 2 L scale was developed. In the shaken scale-down model online monitoring of pH and dissolved oxygen concentration was performed using the Shake Flask Reader (SFR) by PreSens. Though only a simple, manual control of these two parameters could be realized the shaken scale-down model achieved similar data to the stirred-tank model for viable cell density, specific glycosylation content, and ion exchange chromatography pattern. more

New Tricks to Sequence a Protein - Top-Down Mass Spectrometry and Biopharmaceutical Characterization

Proteins become increasingly important in drug development, prompting for their detailed analysis and side product characterization in development and QC. Mass spectrometric Top-Down sequencing (TDS) addresses the key requirement to assign N- and C-terminal sequences in biologics and biosimilars. The principles of the new technology are described and related to the classical Edman-sequencing approach. more

Next-generation Sequencing - Application in Molecular Diagnostics

Next-generation sequencing (NGS) has developed over the past five years into a valuable tool for research applications in the fields of genomics, transcriptomics and epigenomics. Research projects often rely on the analysis of the entire genome. For diagnostic purposes, however, sequencing of the entire human genome is not required and is not even desirable. more

PreSens on Growth Control - Non-invasive Measurement of Dissolved Oxygen & pH in Shake Flask ...

PreSens: Shake flasks are the most commonly used cultivation system. A major, though often neglected problem in aerobic shake flask cultivation is limited oxygen supply or an incorrect pH value. more

Spheroids for Drug Testing - 3D Cell-based Assays

Drug discovery is risky, costly and time consuming. Thus, selection criteria reliably identifying promising drug candidates in early drug development are needed. Because empirical drug testing in simple preclinical models including subcutaneous mice is not sufficient for translation into the clinic, high content (HC) assays reflecting complexity and heterogeneity of patient tumors are required for high quality results. Moreover, these HC models have to be adaptive for high-throughput (HT) drug testing. more

On-line Oxygen Monitoring in Cell Culture

Real-time measurements of oxygen and pH in mammalian cell cultures are important to understand the metabolic dynamics in cell cultures. The observation of the time course of cellular oxygen consumption in response to environmental conditions or toxicological insults can be substantial for successful culture of mammalian cells. This type of monitoring could also be valuable for other applications such as tissue engineering and stem cell research. more
Direct Ethanol Determination

Direct Ethanol Determination

In the field of the biotechnological production processes ethanol can be considered as an end product or an undesired metabolite. In the fermentative production of alcoholic beverages or in the scope of the production of alternative fuels, so-called biofuels, ethanol is a desired end product. In the latter case one aim IS to achieve the highest possible yield of ethanol (space-time yield). more
Low Amount DNA and RNA

Low Amount DNA and RNA

Fluorescent quantification has revolutionized the detection and quantification of small amounts of DNA and RNA. We have developed a fluorescent dye binding assay that allows determination of DNA and RNA from high to low concentrations within a single measuring run sequence, saving time and sample. more
